RESUMO

Objective Study on the intracellular calcium and growth suppression of human breast cancer cells exposed by high intensity ultrasound (HIU). Methods Exposed human breast cancer cells MDA-MB-231 and MCF-7 in vitro with HIU (50 W/cm2). Examined the intracellular calcium from exposed cells with Fura-2 fluorescence probe. The cell viabilities were measured by MTT assay. The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. Results The lever of intracellular calcium went up in pace with exposed time of HIU, MCF-7 cells were (572±20.1), (670±18.9), (815± 16.3) nmol/L (F = 663.65, P<0.001), MDA-MB-231 cell was (582±16.3), (687±19.7), (843± 14.8) nmol/L (F = 863.06, P<0.001), and the distribution of cell cycle waved to G0-G1, the ratios of G0-G1 in MCF-7 and MDA-MB-231 were (60.5±5.5)%, (66.3±7.0)%, (74.5±8.2)% (F=8.17, P = 0.002) and (58.5± 6.3) %, (66.1±6.3)%, (71.2±7.9) % (F=7.51, F= 0.003). Apoptotic rate upgraded gradually, the apoptotic rates of MCF-7 and MDA-MB-231 were (7.3±1.7)%, (13.2±3.5) %, (19.3±3.7)% (F= 18.73, P<0.001) and (6.3±1.8)%, (11.4±2.31)%, (16.4±3.3)% (F = 19.26, P<0.001). Under MTT assay, the rate of cell growth suppression increased significantly, the rates of cell growth suppression in MCF-7 and MDA-MB-231 were (9.2±2.2) %, (24.3±3.9)%, (48.6±5.5)%(F=117.16, P <0.001) and (9.0±1.7)%, (22.3±3.5) %, (416± 6.4)% (F =71.25, P<0.001). Conclusion HIU enhanced the intracellular calcium of human breast cancer cells within given time and promoted the distribution of cell cycle to G0-G1. The rate of cell apoptosis and the cell's death rate increased evidently.

Objective To investigate the expression of snrvivin,an anti-apoptosis gene,in breast cancer tissues and its relationship with clinical parameters of breast cancer patients.To investigate the role of survivin in oncogenesis of breast cancer and its relation with prognosis of breast cancer.Methods Western blot was conducted to detect survivin expression in cancerous tissues and adjacent tissues of 50 breast cancer patients.20 normal breast tissue samples were used as control.Results Survivin was expressed in 38 of the 50 breast cancer tissues (76.0%)and in 11 of 50 breast adjacent normal tissues(22.0%)while no expression was found in normal tissues.The positive protein expression of survivin was related to axillary lymph node metastasis of breast cancer while it was not related to expression of PR,ER,Her-2 or ages of patients.Conclusion Survivin may play a role in breast tumorigenesis and is an important predictive parameter in breast cancer patients.

Objective To investigate the protective effect of heme oxygenase-1 for xeno-heart transplantation and the possible mechanism.Methods Cobalt protoporphyrin (CoPP), HO-1 inducer, and zinc protoporphyrin (ZnPP), HO-1 inhibitor, were used to intervene donors and recipients on the model of NIH-Wistar cardiac transplants respectively and simultaneously some of recipients were treated with immunesuppressor cyclosporine A (CsA), and control group and CsA treated group were set up respectively. The expression of HO-1 protein, HO-1 mRNA, caspase-3 and STAT-3 in transplanted heart tissue was detected by immunohistochemistry, RT-PCR and Western Blot. At the same time, HO-1 enzymatic activity was examined in heart tissue, and the cardiac cell apoptosis was examined by means of TUNEL. Objective To review the mechanisms of breast cancer escaping from host immune surveillance. Methods The current literatures on the mechanisms of breast cancer cells escaping from host immune surveillance were reviewed in the following aspects: alterations of MHC-Ⅰmolecule phenotype, deficiency of costimulatory molecules, apoptosis of T-lymphocytes induced by breast cancer cells presenting Fas ligand (FasL) and TNF-related apoptosis-inducing ligand (TRAIL) and host immune tolerance induced by tumor cells. Results Loss of classical antigen-presenting human leukocyte antigen (HLA) class-Ⅰmolecules, expression of non-classical HLA class-ⅠmoleculesHLA-G, loss of costimulatory molecule B7, apoptosis of T-lymphocytes induced by tumor cells presenting FasL and TRAIL and antigen-inducing host immune tolerance were related to breast cancer escaping from immune surveillance. Conclusion Breast cancer cells escape from host immune surveillance by altering MHC-Ⅰmolecules, lacking of costimulatory molecules, inducing of apoptosis of T cells and host immune tolerance. But the factors resulting in breast cancer escaping from host immune surveillance are not yet clear and should be further studied.

Objective To study the apoptotic induction effect of Thapsigargin on estrogen receptor positive human breast cancer cell lines MCF 7. Methods Cells were treated with Thapsigargin and 5 FU in vitro . The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. The cell viability was measured by MTT assay and ultrastructural changes in apoptotic cells were confirmed by transmission electron microscopy.Results Thapsigargin could increase the rates both of cell apoptosis and growth supression of MCF 7 cells induced by 5 FU and alter the distribution of cell cycle. Under electron microscope, apoptotic bodies in MCF 7 cells considerably increased.Conclusion Thapsigargin apparently enhances the effect of apoptotic induction of 5 FU on MCF 7 cells, it is worthy of being further studied.

Objective To observe the effect of nimesulide (NIM) on DMBA-induced mammary tumors and to investigate possible mechanisms of inhibiting tumors. Methods The Wistar rats were randomly divided into four groups: DMBA group, NIM+DMBA group, NIM+diet group and diet group. The incidence and mean latent phase of mammary tumors were observed. The number and volume of tumors in every rat were measured. RESUMO

Objective To study the liver injury and effects of aescin on liver in rats with acute pancreatitis. Methods The rats were divided into 3 groups (control group, AP group and aescin group). The serum alanine aminotransferase (ALT), serum lactate dehydrogenase (LDH), hepatic cellular energy charge (EC) and adenosine triphosphate (ATP) were detected. The pathologic changes in pancreas and liver were also observed. Results The serum levels of ALT and LDH in aescin group were significantly lower than those of the AP group. The EC and ATP levels were significantly higher in aescin group than that of the AP group. Conclusion Introvenous injection of aescin can alleviate the liver injury in rats with acute pancreatitis.

Objective By reproducing mouse to rat heart xenograft,to determine the effect of leflunomide on NF-?B P65 in the transplanted heart.Methods The recipient rats were randomly divided into 4 groups:group A(n=6)receiving no immunosuppressant as control;group B(n=6)cyclosporine(CsA);group C(n=6)leflunomide(Lef),group D(n=6)Lef plus CsA.The expressions of NF-?B P65 protein and NF-?B P65 DNA binding activity in transplanted heart tissues were determined by immunohistochemistry,Western blot and electrophoretic mobility shift assay(EMSA).Results Administration of CsA alone had no appreciable effect on the survival time of xenografts [(2.50?1.05)d] and could not suppress the protein expression of NF-?B P65 [(263.09?28.81),IOD] and NF-?B P65 DNA binding activity [(227.49?14.83),IOD] in the tissues of xenografts.Administration of Lef alone could significantly prolong the survival time of xenografts [(4.17?1.33)d] and could suppress the protein expression of NF-?B P65 [(173.48?5.85),IOD] and NF-?B P65 DNA binding activity [(109.96?12.46),IOD](P

Objective To explore the mechanism of delayed xenograft rejection(DXR) by dynamic observation of histological and immunohistologic changes in mouse-to-rat cardiac xenografts.Methods A model of mouse-to-rat cardiac heterotopic xenotransplantation in neck was established by cuff technique.NIH mouse hearts not transplanted served as controls(n=4).Some xenograft recipients were killed and cardiac xenografts harvested at end of 3,8,16,24 h(n=4 for each time point) after transplantation.The cardiac hearts(n=16) of some xenograft recipients were not harvested until rejection time to determine their survival time.All heart samples were examined by HE and immunohistochemistry for semi-quantitative determination of antibodies including C3,IgM,IgG,E-selectin and macrophage marker——CD68.Results During the period after transplantation,the degree of rejection of xenografts became more and more serious till ultimate rejection.The mean survival time of the xenografts was(49.3?16.2) h.Immunohistochemical examination showed C3 were not detected in the xenografts at any time during the course of rejection;From 3 h after transplantation,obvious deposition of IgM was found in the grafts and IgG deposition got abundant;E-selectin expression was found as early as 3 h after transplantation and increased gradually;There was progressive infiltration by macrophages in the grafts.Conclusion Mouse-to-rat cardiac xenotransplantation can serve as an animal model of DXR.Endothelial cells activation,IgM and IgG,macrophage infiltration involve in DXR development except C3.

<p><b>OBJECTIVES</b>To observe the expression of somatostatin receptor subtype 2 (SSTR2) mRNA, and investigate the relationship between the expression of SSTR2 mRNA and the expressions of estrogen and progesterone receptors (ERs and PRs) in benign and malignant breast tissues.</p><p><b>METHODS</b>Samples from a total of 23 breast carcinomas, 16 mammary hyperplasias, and 9 mammary fibroadenomas were analyzed. SSTR2 mRNA expression was examined by in situ hybridization using multiphase oligoprobes. ER and PR expressions were detected by immunohistochemical staining. A computerized image analysis system was utilized to estimate the relative content of SSTR2 mRNA.</p><p><b>RESULTS</b>The rate of expression (87.0%) and relative content (0.47) of SSTR2 mRNA in breast cancer were higher than those in benign breast tissue (64%, 0.26) (P < 0.05). SSTR2 mRNA expression was closely correlated with ER and PR expressions in breast cancer (P < 0.05). SSTR2 mRNA was also positively correlated with ER expression in benign breast tissues.</p><p><b>CONCLUSIONS</b>SSTR2 mRNA expression is higher or in benign breast tissues than in malignant ones. There is a significant positive correlation between SSTR2 mRNA and ER and PR expressions. Combined antiestrogen and somatostatin analogue in treatment of ER-positive breast cancers should be further investigated.</p>

Objective To construct the expression vector maspin/pEFIRES-N so as to study maspin gene on the inhibition of the growth,invasion,and metastasis in hepatocellular carcinoma.Methods The full lenth of maspin gene was cloned and ligated to the expression vector pEFIRES-N digested by EcoRⅠ+XbaⅠ by T4 DNA ligase.The recombinant vector maspin/pEFIRES-N was stably transfected into hepatocellular carcinoma MHCC-97 cell line,and the expressive changes of maspin gene were detected.Results The recombinant plasmid was amplified in the E.coli.JM109.After the identification and sequencing,the reconstructive plasmid was confirmed containing the correct and full nucleotide sequence of maspin gene and the mRNA and protein level of maspin gene were up-regulated in hepatocellular carcinoma MHCC-97 cells.The proliferation and invasion of MHCC-97 cells was inhibited.Conclusion The expression vector maspin/pEFIRES-N was constructed successfully and could be expressed in eukaryotic cells.

Objectiver  To establish murine breast precancerous model and evaluate the effect of exogenous estrogen and tamoxifen.  Method  50 female Wistar rats were divided into contol group (n=12), the other 38 received low dose carcinogen (DMBA) by gastrogavage, and then divided into DMBA group (common diet,n=8); DMBA+DES group(DES added chow,n=15); and DMBA+TAM group (TAM added chow,n=15). 3 months later, the status of breast hyperplasia, AgNOR and DNA content were measured.  Results  All indexs in control group and DMBA+TAM group were normal.However,in DMBA group and DMBA+DES group,the breast hyperplasia rate was 75%, 93.3%, AgNOR was 3.71±0.76, 5.60±0.42, DNA was 88.59±7.17, 137.75±3.13 respectively.Compared with control group and DMBA+TAM group, the differences of indexes in DMBA and in DMBA+DES group were significant (P<0.01).  Conclusion  Low dosage DMBA administered by gastrogavage established breast precancerous model. Exogenous estrogen promotes the development of breast precancerous disease induced by DMBA, while TAM suppresses the canceration.

RESUMO

Objective To investigate the diagnosis and treatment of primary retroperitoneal tumor,and improve the early correct diagnostic rate and the resection rate.Method 86 patients with primary retroperitoneal tumor(PRT) were analyzed by review of the clinical materials from 1990 to 2000.Results Abdominal or waist pain was the main symptom,and abdominal or waist mass was the main sign.52.6% percent of the patients got correct diagnosis before operation.62 cases were followed up,the 2-year survival rate for complete resection cases were 61.3%,while the patients who received palliative resection or biopsy died within one year.The resection rate of tumors was 81.3% in benign tumors and 37.7% in malignant tumors,the recurrent rates after complete resection were 15.4%,60%,respectively.Conclusions The earlier diagnosis,totally removal of tumors at first time,periodic examination after operation are three important ways.The operative treatment is very imperative for the patients with recurrence.

Objective To investigate the diagnosis and management of pancreatic injury. Methods The clinical data of 23 patients suffering from pancreatic injury treated by surgery in recent 15 years were analyzed retrospectively. According to the pancreas organ injury scale of the American Association for the Surgery of Trauma, there were 8 cases in grade I, 8 in grade II, 5 in grade III, 1 in grade IV and 1 in grade V. Results Only 2 patients were diagnosed preoperatively out of the 19 patients.About 63.2%(12/19)of the cases had elevated levels of serum amylase. All the patients were cured.In this series,8 cases occurred external pancreatic fistula, 4 pseudocysts, 2 intra-abdominal infections and 1 hyperglycemia postoperatively. Internal pancreatic fistulas were found in all the non operation patients. Conclusions The serum amylase determinations is neither sensitive nor specific for diagnosis of pancreatic injury. Early diagnosis and correct operative pracedure are important to decrease the postoperative complications and improve the outcome.

Objective To investigate the effect of nimesulide(NIM) on human breast cancer cell lines, including MDA?- MB?- 231(estrogen receptor?- negative) and MCF?- 7 (estrogen receptor?- positive). Methods MDA?- MB?- 231 and MCF?- 7 human breast carcinoma cell lines were treated with NIM in vitro. Cell proliferation was evaluated by MTT assay, distribution of cell cycle and rate of apoptosis were determined by flow cytometry, meanwhile the rates of apoptosis were estimated on the basis of Annexin V apoptosis detection. The apoptosis was confirmed by ultrastructural alternations. Results The inhibitory effects of NIM on proliferation of MDA?- MB?- 231 and MCF?- 7 cells were associated with cell cycle arrest in G0/G1 phase and with induction of apoptosis, in a time and dose?- dependent manner. Simultaneously, NIM may also inhibit proliferation and stimulate apoptosis of MCF?- 7 cells. Conclusions NIM inhibits proliferation and induces apoptosis not only in ER?- positive, but also in ER?- negative breast cancer cells. There were two different mechanisms underlying the anti?- carcinogenic action of NIM, namely, dependent on the expression of COX?- 2 protein or not.

Objective To study the effect of rising [Ca 2+ ]i on the expression of Caspase 3, Bax and Bcl 2 in human breast cancer MCF 7 cell lines. Methods After the MCF 7 cells treated with different dose of thapsigargin(TG), intracellular calcium concentrations in MCF 7 cells were measured by calcium sensitive fluorescence indicator Fura 2/AM technique; and immunohistochemical method was used to detected the expressions of Caspase 3, Bax and Bcl 2. Results Bax was over expressed and Bcl 2 was down expressed in a [Ca 2+ ]i concentration dependent way;nevertheless, the expression of Caspase 3 was negative in all the TG treated cells. Conclusions The increase of intracellular calcium concentration may have an inductive effect of apoptosis on MCF 7 cells, which is independent to Caspase 3.

Methods Reveiwing the recently world reference regarding to the diagnosis and treatment of BC was done for making an advancing paper of BC. Results The NF ?B and telomerase may be a new target of anti cancer of breast. The ECT, fiberoptic mammaroductoscopy, examine of micrometastase of BC, immuno cell chemical examine were progressing in the diagnosis of BC. The advance of the treatiment of BC included breast conservative surgery, lymphatic mapping and sentinel node biopsy, biochemical response modifier, new adjuvant chemotherapy, TAM and 1,25 (OH) 2D 3 treatmen. Conclusions The idea and mould of the diagnosis and treatment of BC have advanced obviously.
